A National Geographic crew which included documentary director Terri Flores (Jennifer Lopez), cameraman Danny Rich (Ice Cube) and professor Dr Steven Cale (Eric Stoltz) were shooting a documentary on long lost Indian tribe when they happen to meet a stranded snake hunter Paul Sarone (Jon Voight). They help him believing that he knows about the tribe and he would help them in return. But Paul Sarone takes control of the boat when Dr Steven Cale gets incapacitated due to his scuba gear malfunction. He then starts commanding the crew to help him in achieving his real objective of hunting and capturing the largest snake Anaconda. In this misadventure several members of the crew lose their lives before the snake is finally killed. Paul Sarone then tried to use the two surviving members of the crew, Terri Flores and Danny Rich as a bait to capture the second largest Anaconda but is himself killed along with the snake. To the end of the movie, Terri Flores and Danny Rich are able to locate the lost tribe natives and start filming their documentary.
